Detection of human CD45 by western blot. Samples: Whole cell lysate (50 µg) from Jurkat, MOLT-4, K-562, SR, HeLa, and HEK293T cells prepared using NETN lysis buffer. Antibody: Rabbit anti-CD45 recombinant monoclonal antibody [BL-178-12C7] (A700-012-T lot 1) used at 1:1000. Secondary: HRP-conjugated goat anti-rabbit IgG (A120-101P). Chemiluminescence with an exposure time of 3 seconds.
Detection of human CD45 by western blot of immunoprecipitates. Samples: Whole cell lysate (1 mg per IP reaction; 20% of IP loaded) from Jurkat cells prepared using NETN lysis buffer. Antibodies: Rabbit anti-CD45 recombinant monoclonal antibody [BL-178-12C7] (A700-012-T lot 1) used for IP at 20 µl per reaction. CD45 was also immunoprecipitated by rabbit anti-CD45 antibody (A304-376A). For blotting immunoprecipitated CD45, A700-012-T was used at 1:1000. Chemiluminescence with an exposure time of 3 seconds.
