Detection of human glycosylated and degl
Detection of human glycosylated and deglycosylated UPAR by western blot. Samples: Whole cell lysate (50 µg) from U-937 cells incubated with (+) or without (-) TPA (200nM, 72 hrs) and lysed using NETN lysis buffer. The lysate was then treated (+) or mock treated (-) with PnGase F. Antibodies: Affinity purified rabbit anti-UPAR antibody A304-463A-T (lot A304-463A-T-1) used for WB at 1.0 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.

Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from U-937 cells incubated with (+) or without (-) TPA (200nM, 72 hrs) and lysed using NETN lysis buffer. The lysate was then treated (+) or mock treated (-) with PnGase F. Antibodies: Affinity purified rabbit anti-UPAR antibody A304-462A (lot A304-462A-1) used for IP at 6 µg per reaction. For blotting immunoprecipitated UPAR, A304-463A-T was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.
