Antibody was affinity purified using an epitope from human PKD2 immobilized on solid support. The epitope recognized by A300-073A maps to a region between residues 320 and 400 using the numbering given in entry NP_057541.2 (GeneID 25865). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human PKD2 by western blot. Samples: Whole cell lysate (50 µg) from HeLa and HEK293T cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-PKD2 antibody A300-073A (lot A300-073A-2) used for WB at 0.4 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.
Detection of human PKD2 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-PKD2 antibody A300-073A (lot A300-073A-2) used for IP at 6 µg per reaction. PKD2 was also immunoprecipitated by a previous lot of this antibody (lot A300-073A-1) and rabbit anti-PKD2 antibody A300-074A For blotting immunoprecipitated PKD2, A300-073A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.
