Antibody was affinity purified using an epitope specific to CD3E immobilized on solid support. The epitope recognized by A304-291A maps to a region between residue 157 to 207 of human CD3e Molecule, Epsilon (CD3-TCR Complex) using the numbering given in entry NP_000724.1 (GeneID 916). Antibody concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human CD3E by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from Jurkat cells. Antibodies: Affinity purified rabbit anti-CD3E antibody A304-291A (lot A304-291A-1) used for IP at 6 µg per reaction. CD3E was also immunoprecipitated by rabbit anti-CD3E antibody BL16032 and rabbit anti-CD3E recombinant monoclonal antibody [BL-298-5D12] (A700-016). For blotting immunoprecipitated CD3E, A700-016 was used at 1:1000. Chemiluminescence with an exposure time of 10 seconds.
