Palladin, cytoskeletal associated protein (PALLD) is a cytoskeletal protein crucial for organizing the actin cytoskeleton, influencing cell shape, motility, and adhesion. It exists in various isoforms due to alternative splicing and is predominantly expressed in mesenchymal cells. Mutations in PALLD are linked to familial pancreatic cancer, with specific mutations like P239S associated with increased cell migration and invasive potential in cancer cells. Overexpression of palladin has also been observed in pancreatic neoplasia, suggesting its role in tumor microenvironment alterations rather than directly within cancer cells themselves. Additionally, PALLD mutations have been implicated in other diseases, including dilated cardiomyopathy and myocardial infarction, highlighting its significance beyond cancer biology.
