Lysine demethylase 3A (KDM3A), also known as JMJD1A, is a histone demethylase that specifically targets mono- and dimethylated lysine 9 on histone H3, playing a crucial role in gene expression regulation and transcriptional activation, particularly in hormone-dependent pathways. KDM3A is implicated in various biological processes, including spermatogenesis and metabolic regulation, contributing to obesity resistance by modulating genes such as PPARα and UCP1. Dysregulation of KDM3A has been associated with several diseases, notably cancer; it facilitates chemoresistance and cancer stemness by demethylating key proteins like p53, thereby inhibiting apoptotic pathways. Additionally, KDM3A's overexpression is linked to prostate cancer progression through its interaction with androgen receptors and hypoxia-inducible factors, making it a potential therapeutic target for cancer treatment.
