WWP2 (WW Domain Containing E3 Ubiquitin Protein Ligase 2) is an E3 ubiquitin ligase that plays a crucial role in various biological processes, including protein ubiquitination, chondrogenesis, and oncogenic signaling pathways. It interacts with Smad proteins and the tumor suppressor PTEN, influencing cell signaling and differentiation. WWP2 has been linked to several diseases, including Spondyloepiphyseal Dysplasia, Nishimura Type, and Cowden Syndrome 1, and its dysregulation can contribute to fibrosis, inflammation, and cancer progression. The inhibition of WWP2 has been identified as a potential therapeutic strategy for treating diseases characterized by fibrosis and inflammation, as it regulates the expression of pro-inflammatory factors and fibroblast activation. Additionally, WWP2 isoforms have distinct functions, with WWP2-N potentially suppressing TGFβ-induced epithelial-to-mesenchymal transition (EMT) and WWP2-C interacting with inhibitory Smad7.
