Microfibrillar-associated protein 5 (MFAP5), also known as microfibril-associated glycoprotein 2 (MAGP2), is an extracellular matrix protein that plays a significant role in the structural integrity and function of microfibrils. It is primarily involved in the assembly and stabilization of elastic fibers, contributing to tissue elasticity and resilience. MFAP5 interacts with various cellular components, including integrins such as αvβ3, promoting cell adhesion and migration. This protein is particularly important in wound healing and tissue repair, where it regulates fibroblast behavior, collagen deposition, and angiogenesis. Dysregulation of MFAP5 has been implicated in several diseases, including idiopathic pulmonary fibrosis and various cancers, such as basal-like breast cancer, where its overexpression correlates with enhanced tumor progression and metastasis.
