BioSkryb’s ResolveDNA® kit is designed to address the challenges of DNA analysis at a single-cell level with unmatched accuracy and efficiency. ResolveDNA enables researchers to generate high-quality genomic data, even from minimal starting material. This is due to its primary template amplification (PTA) technology for whole genome amplification (WGA), which ensures consistent and comprehensive amplification across the genome, reducing gaps and increasing reliability for downstream analyses. This method also reduces amplification artefacts, enabling accurate detection of single-nucleotide variants (SNVs) and copy number variants (CNVs). It provides the precision necessary to identify rare mutations and analyse genetic heterogeneity at a single-cell level.

Primary template amplification technology
PTA can be performed using single cells (obtained via FACS, microfluidics, or similar methods), multiple cells, or ultra-low DNA inputs ranging from >4 pg to 10 ng. Following denaturation, random primers (6- to 9-mers) are annealed. Amplification with phi29 polymerase and a specialized nucleotide pool produces amplicons ranging from approximately 250 bp to over 1,500 bp. The relatively short size of these amplicons promotes re-priming on the original template, minimizing the exponential amplification of biases and errors in the resulting molecules. Additionally, PTA chemistry effectively reduces the generation of artifacts such as chimeric molecules and non-specific priming. The double-stranded PTA products can then be prepared for library construction and multiplexed sequencing on platforms like Illumina® or others.
