The MosaiX™ DNA Library Preparation Kit from SeqWell is a fast, simple, and scalable solution for NGS library preparation, designed to meet the demands of modern genomics. Built on advanced directional tagmentation using TnX transposase, MosaiX delivers low bias tagmentation with reduced TnX insertion bias, enabling the construction of high complexity DNA libraries with a low duplication rate and uniform genome coverage. It is ideal for whole genome sequencing, whole exome sequencing, and population genomics.
This 90-minute library prep kit offers a powerful ligation-based alternative, streamlining high throughput DNA sequencing prep workflows. Suitable for library prep for human genome sequencing and other large-scale applications, MosaiX provides a robust tagmentation-based library prep approach.
Benefits of MosaiX
- Fast, simplified workflow: complete in 90 minutes, with only 35 minutes hands-on time, speeding up sample preparation.
- High library complexity: produces high-complexity, low-duplication libraries ideal for NGS applications.
- Uniform sequence coverage: ensures consistent and accurate genome coverage using directional tagmentation.
- Scalable for high-throughput sequencing: designed for population genomics and large-scale sequencing projects.
- Reliable performance: low bias and consistent results, even for complex genomic studies.
Reduced insertion bias - the TnX difference
TnX is an engineered transposase optimised to minimise sequence-dependent insertion preferences. This results in libraries with improved complexity, better uniformity, and more accurate downstream sequencing data, especially beneficial for applications requiring high fidelity and reduced bias. The above image shows positions with higher per-base nucleotide bias represented by heights for hyperactive Tn5 and TnX, and illustrates the reduced bias of TnX.
Applications of MosaiX
- Whole genome sequencing: directional tagmentation followed by ligation supports the preparation of the complex libraries needed for WGS
- Population genomics: efficient for sequencing large populations with high complexity
- Microbial genomics: scalable, plate-based workflows enable high-throughput sequencing of microbial genomes
- Targeted sequencing: enzymatic fragmentation produces consistent, appropriately sized fragments ideal for hybrid capture or amplicon-based enrichment
- Metagenomics: TnX’s reduced insertion bias results in uniform coverage across a range of microbial genomes, reducing GC bias and ensuring low-abundance species are not missed
