Cross-adsorbed secondary antibodies

High-specificity antibodies for multiplex and low background imaging

Cross-adsorbed secondary antibodies from Jackson ImmunoResearch are designed to minimise unintended off-target binding (species cross-reactivity) and improve signal specificity in demanding immunodetection workflows. By removing unwanted reactivity against serum proteins and immunoglobulins from non-target species, these highly refined antibodies enable cleaner staining, more reliable multiplexing, and superior experimental reproducibility across research and diagnostic applications.

Ideal for immunofluorescence, flow cytometry, western blotting, ELISA, and immunohistochemistry, cross-adsorbed antibodies help researchers achieve strong signal intensity while significantly reducing background interference and non-specific binding.

Key features

  • Reduces non-specific background staining: caused by endogenous tissue immunoglobulins and Fc-mediated binding
  • Improve multiplex performance: in the presence of cross-reacting immunoglobulins and improve channel separation
  • Enhanced signal specificity: reduced off-target binding and cleaner fluorescent and chromogenic signals
  • Reliable performance: IHC, IF, ICC, ELISA, western blotting and flow cytometry

What are cross-adsorbed secondary antibodies?

Secondary antibodies naturally recognise immunoglobulins from related species. Without adsorption, this can lead to unwanted cross-reactivity, background staining, and false-positive signals in complex experiments.

Cross-adsorbed secondary antibodies are affinity-purified antibodies that have been further adsorbed against serum proteins or immunoglobulins from additional species. This process removes antibodies with undesirable binding activity while retaining strong affinity for the intended target species. Depending on the antibody description, the gels used for cross-adsorption may contain proteins from different species, different mouse IgG subclasses, or different antibody fragments. Jackson ImmunoResearch confirms that species cross-reactivity has been minimized by ELISA and IEP testing.

Process of creating cross-adsorbed antibodies

Creation of cross-adsorbed antibodies

Products labelled with “min X … Sr Prot” indicate minimal cross-reactivity against the listed species’ serum proteins and immunoglobulins, helping improve specificity in multi-species experiments.

The following abbreviations are used in the parentheses:

min X = minimal cross-reaction  Ar Hms = Armenian Hamster Rb = Rabbit
Bov = Bovine Sy Hms = Syrian Hamster Shp = Sheep
Ck = Chicken Hrs = Horse Sw = Swine
Gt = Goat Hu = Human Sr = Serum
GP = Guinea Pig Ms = Mouse Prot = Protein

Cross- adsorption and multiplex labelling

In multiple-labelling experiments, secondary antibodies may unintentionally recognise other primary antibodies or endogenous proteins if they are not sufficiently adsorbed.

Highly cross-adsorbed secondary antibodies are specifically designed to overcome this challenge by minimising binding to serum proteins from related species.

Typical multiplex workflow benefits

  • Simultaneous detection of multiple biomarkers
  • Reduced false-positive staining
  • Cleaner spectral separation
  • Improved reproducibility

Jackson ImmunoResearch highly cross-adsorbed antibodies are widely used in advanced imaging workflows and multiplex fluorescence applications.

Choosing the right cross- adsorbed secondary antibody

Selecting the correct secondary antibody depends on your sample type, primary antibody species, and detection workflow
