ChIP-validated antibodies

Chromatin immunoprecipitation (chromatin IP or ChIP) is an extremely challenging technique which requires the use of high-quality antibodies. For an antibody to work in chromatin IP, it must be of high titre and very specific, with no detection of non-target proteins. Most importantly, it must be able to recognise the target protein in its native chromatin-associated context after fixation.

Active Motif specialises in manufacturing high-quality antibodies to histones, histone modifications and chromatin proteins, and validating them for use in ChIP, as well as ChIP-Seq and ChIP-chip.

Histone H3 acetyl Lys27 antibody tested by ChIP qPCR analysis

Histone H3 acetyl Lys27 antibody tested by ChIP qPCR analysis. ChIP was performed using the ChIP-IT® High Sensitivity Kit with chromatin from a human hepatocyte cell line. qPCR was performed using a negative control primer set (Untr20) and three positive control primer sets that amplify known H3K27Ac sites.

Histone H3 trimethyl Lys27 pAb tested by ChIP-Seq

Histone H3 trimethyl Lys27 pAb tested by ChIP-Seq. ChIP was performed using chromatin from the human iPS 19.11 cell line. ChIP DNA was sequenced on the Illumina® GA II and sequence tags were mapped to identify H3K27me3 binding. The image shows H3K27me3 binding across a 140,000 bp region of the HOX gene cluster on chromosome 12.

5-Hydroxymethylcytosine (5-hmC, 5-hydroxymethylcytidine) antibody tested by Methyl DNA immunoprecipitation

5-Hydroxymethylcytosine (5-hmC, 5-hydroxymethylcytidine) antibody tested by Methyl DNA immunoprecipitation. DNA (25 pg) derived from the promoter of the APC gene was spiked into 500 ng of human genomic DNA and subjected to the MeDIP procedure using 1 μl of 5-Hydroxymethylcytidine antibody (5hmC, maroon bars) or 1 μl of control rabbit IgG (IgG, blue bars). Real time quantitative PCR was performed on the immunoprecipitated DNA and results plotted as % of input DNA. The spiked APC DNA contained either no methylation (DNA), 5-methylcytosine methylation (5-mC) or 5-hydroxymethylcytosine methylation (5-hmC).
