T-cell receptors (TCRs) are vital to the adaptive immune system, recognising specific antigen peptides presented by MHC molecules on the surface of infected or abnormal cells. TCRs are essential for initiating immune responses against pathogens or abnormal cells, such as cancer cells. Sequencing these antigen-specific TCRs has made it possible to engineer TCRs into T cells. Using this method, T cells are modified and expanded before infusion back into the patient to target and eliminate cancer cells that express the specific antigen.
TCR cell therapy research tools from BPS Bioscience
BPS Bioscience offer a range of innovative solutions to your cell therapy needs when it comes to using TCRs. The range is designed for targeting of NY-ESO-1 (New York esophageal squamous cell carcinoma) and MART-1 (Melanoma-associated antigen recognised by T cells).
- Reporter cells
TCR-expressing reporter cells are available in ready-to-use format to aid in the design and optimisation of co-culture bioassays or to serve as positive controls in these assays.
BPS offer TCR knockout reporter cell lines which enable researchers to reintroduce specific TCR variants, allowing for the comparison of how different TCRs affect signaling pathways.

- Lentiviruses
BPS Bioscience offer ready-to-use lentiviruses efficiently transduce immune cells with antigen-specific TCRs to create either transient or stable cell lines. These high-titer lentiviral particles are provided pre-prepared - simply add them to your cells and optionally select with puromycin.
T cell activation using T2 cells as APC

CD8+ TCR knockout/NFAT Luciferase Reporter Jurkat cells (#78757) were transduced with lentiviruses expressing different TCR variants via spinoculation at an MOI of 10. After 66 hours, the cells were co-cultured with T2 cells loaded with either NY-ESO-1 peptide (#78758) or MART-1 peptide (#78760) for 6 hours. A luciferase assay was then conducted, and the results are presented as raw luminescence readings.
- Peptides
Native and mutated NY-ESO-1 and MART-1 antigen peptides can be utilised in competition assays or for loading antigen-presenting cells in co-culture bioassays.
