Antibody was affinity purified using an epitope specific to TRIP4/ASC-1 immobilized on solid support. The epitope recognized by A300-203A maps to a region between residues 75 and 125 of human thyroid hormone receptor interactor 4,(activating signal cointegrator 1) using the number given in entry NP_057297.2 (GeneID 9325).Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human TRIP4/ASC1 by western blot. Samples: Whole cell lysate (50 µg) from HeLa and HEK293T cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-TRIP4/ASC1 antibody A300-203A (lot A300-203A-1) used for WB at 0.1 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.

Detection of human TRIP4/ASC1 by western blot and immunoprecipitation. Samples: Nuclear extract (5 µg for WB; 1 mg for IP) from HeLa cells. Antibodies: Affinity purified rabbit anti-ASC1 antibody (Cat. No. A300-203A) used at 0.3 µg/ml for WB and 4 µg/mg lysate for IP. immunoprecipitatesd ASC1 was detected using an antibody (BL458) to a distinctly different epitope on ASC1. Detection: A) An alkaline phosphatase conjugated secondary antibody with NBT-BCIP development for 20 minutes. B) An HRP conjugated secondary antibody with chemiluminescent exposure for 4 minute.
