Detection of human GR by western blot and immunoprecipitation. Samples: Whole cell lysate from HeLa (15 and 50 µg for WB; 1 mg for IP, 20% of IP loaded) and HEK293T (T; 50 µg) cells. Antibodies: Affinity purified rabbit anti-GR antibody A303-491A used for WB at 0.04 µg/ml (A) and 0.4 µg/ml (B) and used for IP at 6 µg/mg lysate. GR was also immunoprecipitated by rabbit anti-GR antibody A303-490A, which recognizes an upstream epitope. Detection: Chemiluminescence with exposure times of 30 seconds (A) and 1 second (B).
Localization of GR Binding Sites by ChIP-sequencing. Chromatin from prednisolone treated THP-1 cells was immunoprecipitated with anti-GR antibody A303-491A and analyzed by DNA sequencing. The figure illustrates the peak distribution of GR binding within a 250 Kb region of chromosome 6 as detected using anti-GR A303-491A. ChIP-seq validation performed by Active Motif, Carlsbad, CA.
