The Exo-Fect™ reagent enables the high efficiency transfection of nucleic acids directly into isolated exosomes. In less than one hour, create novel exosome delivery vehicles for siRNA, miRNA, mRNA, plasmid DNA and even small molecules for transport into target cells.
Benefits of using Exo-Fect
• Simple protocol
• No electroporation required
• Load siRNA, miRNA, mRNA and plasmid DNA
• Load small molecules for drug discovery applications
• Texas Red-labelled positive control included
Exo-Fect efficiently loads small RNAs into exosomes for delivery to recipient cells
Fluorescence imaging shows exosomes that were loaded with either a Texas Red end-labelled siRNA (left panels), or unlabelled siRNA (right panels) using Exo-Fect, and then immobilised onto Exo-Flow CD63 magnetic beads.
Exo-Fect transfected exosomes deliver siRNA cargo to recipient cells.
Transfection of mRNAs and plasmid DNA directly into exosomes
Left panels: 1 µg of mRNA encoding RFP was transfected into exosomes. These exosomes were added to HEK293 cells and imaged for RFP protein production after 24 h. Right panels: 5 µg of plasmid DNA encoding a GFP gene was transfected into exosomes and added to HEK293 cells. The cells were imaged for GFP protein presence after 48 h.
Working with hard-to-transfect cells?
We also offer an easy-to-use exosome-based kit for the delivery of nucleic acids including siRNA, miRNAs, mRNAs & plasmid DNA to a variety of hard-to-transfect cells. Find out more.
