Detection of human p53 by western blot. Samples: Whole cell lysate (15 µg) from Hep-G2, A-549, and Jurkat cells prepared using NETN lysis buffer. Antibody: Mouse monoclonal anti-p53 antibody [DO-1] (A500-015A-T lot 2) used for WB at 1:1000. Secondary: HRP-conjugated goat anti-mouse IgG (A90-116P). Chemiluminescence with an exposure time of 3 minutes.

Detection of human p53 by western blot of immunoprecipitates. Samples: Whole cell lysate (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from 293T cells. Antibodies: Mouse monoclonal anti-p53 antibody [DO-1] (A500-015A-T lot 2) used for IP at 20 µl per reaction. p53 was also immunoprecipitated by rabbit anti-p53 antibody (A300-247A). For blotting immunoprecipitated p53, A500-015A-T was used at 1:1000. Secondary: HRP-conjugated goat anti-mouse IgG (A90-116P). Chemiluminescence with an exposure time of 3 minutes.
