Antibody was affinity purified using an epitope specific to ADAMTS13 immobilized on solid support. The epitope recognized by A301-000A maps to a region between residue 75 and 125 of human ADAM (a disintegrin-like and metalloprotease) metallopeptidase with thrombospondin type 1 motif, 13 using the numbering given in entry NP_620594.1 (GeneID 11093). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human ADAMTS13 by western blot and immunoprecipitation. Samples: Pooled normal human serum (300 µl/IP, 20% of IP loaded; 1.5 µl for input). Antibodies: Agarose immobilized goat anti-ADAMTS13 antibodies S300-391, S301-000, S301-001, S301-002 and S301-003 were used at the equivalent of 35 µg of antibody per IP. For blotting immunoprecipitated ADAMTS13, goat anti-ADAMTS13 antibody A300-002A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds. Note: The anti-ADAMTS13 antibodies A300-391A, A301-000A, A301-001A, A301-002A and A300-003A where conjugated to agarose to produce the agarose immobilized antibodies S300-391, S301-000, S301-001, S301-002 and S301-003, respectively.
