Antibody was affinity purified using an epitope specific to LEDGF/p75 immobilized on solid support. The epitope recognized by A300-847A maps to a region between residue 225 and 275 of human Lens Epithelium Derived Growth Factor/Transcriptional Coactivator p75 using the numbering given in entry NP_150091.2 (GeneID 11168). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human LEDGF/p75 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-LEDGF/p75 antibody A300-847A (lot A300-847A-3) used for IP at 3 µg per reaction. LEDGF/p75 was also immunoprecipitated by rabbit anti-LEDGF/p75 antibody A300-848A. For blotting immunoprecipitated LEDGF-p75, A300-847A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 75 seconds.
