Antibody was affinity purified using an epitope specific to MCAK immobilized on solid support. The epitope recognized by A300-806A maps to a region between residue 50 and 100 of human Mitotic Centromere-Associated Kinesin using the numbering given in entry NP_006836.1 (GeneID 11004). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human MCAK by western blot of immunoprecipitates. Samples: Whole cell lysate (1 mg for IP, 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-MCAK antibodies BL3505 (Cat. No. A300-806A), BL3507 (Cat. No. A300-807A), and BL3508 (Cat. No. A300-808A) used for IP at 3 µg/mg lysate. For blotting immunoprecipitated MCAK, BL3507 (left panel) and BL3508 (right panel) were used at 1 µg/ml. Detection: Chemiluminescence with exposure times of 10 seconds (left) and 1 second (right).
