Antibody was affinity purified using an epitope specific to ErbB4 immobilized on solid support. The epitope recognized by A300-666A maps to a region between residues 1100 and 1150 of human v-erb-a erythroblastic leukemia viral oncogene homolog 4 using the numbering given in entry NP_005226.1 (GeneID 2066). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human ErbB4 by western blot of immunoprecipitates. Samples: Whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) from HeLa cells prepared using NETN lysis buffer. Antibodies: Affinity purified rabbit anti-ErbB4 antibody A300-666A (lot A300-666A-2) used for IP at 3 µg per reaction. ErbB4 was also immunoprecipitated by rabbit anti-ErbB4 antibody BL3041. For blotting immunoprecipitated ErbB4, A300-666A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.
