Antibody was affinity purified using an epitope specific to PRKRIR immobilized on solid support. The epitope recognized by A300-586A maps to a region between residues 150 and 200 of human Protein-Kinase, Interferon-inducible double strand RNA dependent Inhibitor, Repressor of (P58 Repressor) using the numbering given in entry NP_004696.2 (GeneID 5612). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of PRKRIR by western blot and immunoprecipitation. Samples: Whole cell lysate from HeLa (5, 15 and 50 µg for WB; 1 mg for IP; 1/4 of IP reaction loaded) and mouse NIH 3T3 (M; 50 µg for WB) cells. Antibodies: Affinity purified rabbit anti-PRKRIR antibody A300-586A (lot A300-586A-1) used for WB at 0.04 µg/ml (A) and 1 µg/ml (B) and used for IP at 3 µg/mg lysate (B). PRKRIR was also immunoprecipitated using rabbit anti-PRKRIR antibody BL2254 at 3 µg/mg lysate. Detection: Chemiluminescence with exposure times of 30 seconds (A and B).
