Antibody was affinity purified using an epitope specific to CARM1 immobilized on solid support. The epitope recognized by A300-420A maps to a region between residues 450 and 500 of human Coactivator-Associated Arginine Methyltransferase 1 using the numbering given in Swiss-Prot entry Q86X55 (GeneID 10498). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human and mouse CARM1 by western blot. Samples: Whole cell lysate (50 µg) from HeLa and mouse NIH 3T3 cells prepared using NETN lysis buffer. Antibody: Affinity purified rabbit anti-CARM1 antibody A300-420A (lot A300-420A-1) used for WB at 0.06 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.
Detection of human CARM1 by western blot and immunoprecipitation. Sample: Whole cell lysate (25 µg for WB in A; 5 mg for IP in B) from HeLa cells. Antibody: Affinity purified rabbit anti-CARM1 antibody (Cat. No. A300-420A) used at 0.1 µg/ml for WB and at 4 µg/5 mg lysate for IP. Detection: Chemiluminescence with an exposure time of 1 minute in A. Coomassie stain in B. The identity of the band that is CARM1 was verified by mass spectrometry.
