Antibody was affinity purified using an epitope specific to BLM immobilized on solid support. The epitope recognized by A300-110A maps to a region between residue 1375 and the C-terminus (residue 1417) of human Bloom Syndrome using the numbering given in entry NP_000048.1 (GeneID 641). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human BLM by western blot of immunoprecipitates. Samples: Whole cell lysate (1 mg for IP; 20% of IP loaded) from HeLa cells. Antibodies: Affinity purified rabbit anti-BLM antibody A300-110A (lot A300-110A-3) used for IP at 6 µg/mg lysate. BLM was also immunoprecipitated by a previous lot of this antibody (lot A300-110A-2) and A300-120A. For blotting immunoprecipitated BLM, A300-110A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 30 seconds.
