Antibody was affinity purified using an epitope specific to CDC25a immobilized on solid support. The epitope recognized by A300-075A maps to a region between residues 175 and 225 of human Cell Division Cycle 25A using the numbering given in entry NP_001780.2 (GeneID 993). Immunoglobulin concentration was determined by extinction coefficient: absorbance at 280 nm of 1.4 equals 1.0 mg of IgG.

Detection of human CDC25a by western blot. Samples: Nuclear extract (50 µg) from HeLa and HEK293T cells. Antibody: Affinity purified rabbit anti-CDC25a antibody A300-075A (lot A300-075A-2) used for WB at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.
Detection of human CDC25a by western blot of immunoprecipitates. Samples: Nuclear Extract (0.5 or 1.0 mg per IP reaction; 20% of IP loaded) from HEK293T cells. Antibodies: Affinity purified rabbit anti-CDC25a antibody A300-075A (lot A300-075A-2) used for IP at 6 µg per reaction. CDC25a was also immunoprecipitated by rabbit anti-CDC25a antibody A300-076A. For blotting immunoprecipitated CDC25a, A300-075A was used at 1 µg/ml. Detection: Chemiluminescence with an exposure time of 3 minutes.
