Gibson Assembly® Kits

Cambridge Bioscience offers a range of kits using the Gibson Assembly® method for efficient and simple DNA cloning or for the creation of plasmid constructs.

What is Gibson Assembly®?
An highly efficient technique for rapid assembly of multiple DNA fragments at one time.

Why was Gibson Assembly® developed?
It was developed in 2009 by Dr Daniel Gibson and his colleagues at J. Craig Venter Institute (JCVI) in collaboration with Synthetic Genomics, Inc (SGI), during the team’s quest to construct the first synthetic cell. Since its introduction, the Gibson Assembly® method has become a mainstay in many synthetic biology labs and has attracted interest from both the academic and commercial life sciences community due to its ease-of-use, robustness and flexibility.

What makes Gibson Assembly® advantageous to labs cloning DNA or creating plasmid constructs?
• No need for specific restriction sites
• Join almost any two fragments regardless of sequence
• No scar between joined fragments
• Fewer steps compared to traditional cloning
• Can combine multiple DNA fragments at once

Kits available
For the creation of plasmid clones, Cambridge Bioscience offer two formats:
1. Gibson Assembly® Ultra Kits and Master Mixes
Optimised for the assembly of up to 15 DNA fragments from 100 bp to 100 kb in length, the Gibson Assembly® Ultra kit requires only nanogram amounts of DNA. Using any vector and with no requirements for restriction enzyme sites, the two-step reaction takes only 80 minutes with less than six minutes hands-on time involved.
2.Gibson Assembly® HiFi 1-Step Kits and Master Mixes
One-tube, one-step isothermal method for the creation of large recombinant DNA constructs with up to five inserts in only one hour.

3. Gibson Assembly® Site Directed Mutagenesis Kits
Designed is a two-in-one product, this kit not only has reagents that allow incorporation of up to five different mutations (such as base pair changes, insertions and deletions) but also includes the Gibson reagents to allow assembly of these fragments into the final construct.
