RNA library preparation for transcriptomics

Leveraging Swift Biosciences’ innovative and patented Adaptase® technology, the Swift RNA library kits enable stranded RNA library construction directly from 1st strand cDNA, without the requirement for 2nd strand cDNA synthesis and degradation, or template-switching methods. Compatible with manual and automated workflows as well as upstream and downstream enrichment and depletion methods, the Swift RNA library kits support a variety of indexing options.

Swift RNA library kit
The Swift RNA library kit offers a fast NGS transcriptomics workflow for Illumina® sequencing platforms. With a wide range of input amounts, Swift’s Adaptase technology allows for minimal adapter dimers with no need for adapter titration resulting in <1% adapter dimers and maintenance of ligation efficiency at all supported input levels.

10 ng Universal Human Reference Total RNA (Agilent® 740000) was poly(A) enriched using the NEBNEXT® poly(A) mRNA Magnetic Isolation Module (NEB® E7490) before being processed through the Swift RNA Library Kit or processed following the supplier recommendations. Libraries were sequenced on a MiniSeq™ with 2 x 75 paired-end reads. Fastq files were down sampled to 2.2 million reads before analysis using STAR mapping rate (A) or rnaseqc - genes detected (B), transcripts detected (C), duplication rate (D).

Enabling stranded RNA library construction directly from 1st strand cDNA, the Swift rapid RNA library kit speeds up library preparation allowing researchers to process more samples to increase data output.

Universal Human Reference (UHR) Total RNA (Agilent® 740000) was poly(A)-enriched using the NEBNEXT® poly(A) mRNA Magnetic Isolation Module (NEB® E7490) before being processed through the Swift Rapid RNA Library Kit or processed following the supplier recommendations. Libraries were sequenced on a MiniSeq™ with 2 x 75 paired-end reads. Fastq files were downsampled to 3.8 million reads before analysis using STAR (mapping rate) or rnaseqc (strand specificity).
