Cumate inducible lentivectors

The SparQ™ Cumate Switch Lentivectors enable inducible cDNA or microRNA expression in mammalian cells. Working through virus transduction, these cumate inducible lentivectors deliver extremely tight control, robust induction and a highly titratable expression switch for inducible gene and microRNA expression studies.

The cumate switch inducible system
The cumate switch inducible system works through the CymR repressor that binds the cumate operator sequences (CuO) with high affinity. The repression is alleviated through the addition of Cumate, a nontoxic small molecule that binds to CymR. This system has a dynamic inducibility, can be finely tuned and is reversible and inducible over and over for timed expression studies.

Vector formats available
We are pleased to offer the following vector formats:

1. SparQ All-in-one Cumate Switch Vectors
A simple way to make stable cell lines (using the Puro) marker and to turn on the switch by adding cumate. The inserted gene or microRNA at the MCS is expressed upon induction (QM800A-1) or coinduction with IRES-mediated GFP marker to monitor induction when the switch is turned on (QM812B-1). The cDNA insert size limit is up to 2kb in size.

2. SparQ IRES Vectors
The SparQ™ IRES vector configuration enables the inducible expression of a microRNA or a cDNA along with an IRES-mediated GFP or RFP marker that wil be coexpressed to monitor induction when the switch is turned on.

3. SparQ™ T2A Vectors
The SparQ™ T2A vector configuration enables the inducible expression of a cDNA along with the simultaneous, T2A-mediated GFP or RFP marker to monitor cumate switch induction.

4. SparQ™ Dual Promoter Vectors
The SparQ™ dual promoter vector configuration enables the inducible expression of a microRNA or a cDNA. The downstream EF1 promoter will constitutively express GFP (QM511B-1) or RFP (QM512B-1), or the GFP+Puro (QM513B-1), RFP+Puro (QM516B-1) markers independent of the cumate switch activation.
