COVID sequencing

The SARS-CoV-2 Swift Normalase® Amplicon Panels (SNAP) for next-generation sequencing provide full SARS-CoV-2 genome coverage (99.7%) to enable the identification of SARS-CoV-2 strains, including variants, from a wide range of samples including nasopharyngeal or oropharyngeal swabs, sputa, bronchoalveolar lavage (BAL) and stool. These panels use overlapping primers to generate 345 amplicons (average 150 bp) along the length of the viral genome, ensuring that novel variants are detected, even when the mutation is a dropout of genomic sequence.

Complete genome coverage enables detection of key variants
The SNAP SARS-CoV-2 kit uses a single-tube multiplex primer pool to general amplicons along the length of the 29.9 kb viral genome. Sequencing the full-lenght SARS-CoV-2 genome is increasingly more important to enable detection of known variants, as well as the discovery of novel mutations. The SNAP SARS-CoV-2 kit enables confident mutation detection for tracking nucleotide variants and improving out understanding of virus evolution, transmission and pathogenesis.

Complete genome coverage enables detection of key variants

A nasopharyngeal swab specimen produced with the SNAP SARS-CoV-2 kit and sequencing with Illumina® MiSeq® to 50,000 reads demonstrates coverage across the 29.9 kb genome. The A23403G/D614G mutation was detected in this sample, which affects the viral spike protein and is a key variant of interest for its hypothesised role in transmissibility of COVID-19.

High performance over a wide range of viral copy number

SARS-CoV-2 synthetic template material was mixed with UHR RNA and converted into first-strand cDNA using the SuperScript® IV First-Strand Synthesis System. cDNA was processed with the Swift SNAP SARS-CoV-2 kit, sequencing with Illumina® MiSeq® 2 x 150 bp chemistry and downsampled to 50,000 or 1 million reads per sample.
