miR-ID® microRNA Detection Platform

SomaGenics assays enable sensitive and highly specific quantification of low abundance small RNAs, such as microRNAs, from total RNA isolates. With higher specificity, sensitivity and versatility than other methods, the miR-ID® microRNA Detection Platform is the only qPCR-based platform able to distinguish small RNAs with commonly occurring end modifications such as 5' triphosphates,  5’ OH(e.g., secondary siRNAs in C. elegans), and 2'-OMe modifications (e.g. 3' ends of piRNAs in animals and all small RNAs in plants). Specifically, miRNA modifications (isoforms/isomiRs) and miRNAs with single nucleotide polymorphisms can be distinguished by miR-ID®.

• Quantitatively discriminate between small RNAs with an identical sequence but differing terminal modifications

Step 1: Circularisation of a linear miRNA; Step 2: Reverse transcription mediated by rolling circle amplification to obtain multimeric cDNA; Step3: Quantification of the amplicon by RT-qPCR using highly sequence-specific 5’-overlapping primers and SYBR Green detection.
