1. AGK2
AGK2 is a potent inhibitor of SIRT2 (IC50 = 3.5µM). AGK2 is over 15-fold more selective for SIRT2 than SIRT1 and SIRT3.

2. Cambinol
Cambinol is an inhibitor of NAD-dependent deacetylase activity of human SIRT1 (IC50 = 56µM) and SIRT2 (IC50 = 59µM). The treatment of BCL6-expressing Burkitt lymphoma cells with Cambinol induces apoptosis accompanied by hyperacetylation of BCL6 and p53.

3. EX-527
EX-527 is a cell-permeable, selective inhibitor of SIRT1 (IC50 = 98nM). EX-527 has been used to investigate the relationship between SIRT1-mediated deacetylation of p53, p53 activity, and cell survival following DNA damage. Ex-527 can also inhibit other SIRTs at much higher concentrations and has no effect on other HDACs.

4. Salermide
Salermide is a cell permeable inhibitor of SIRT1 and SIRT2. Salermide has been shown to have a strong pro-apoptotic effect on MOLT4 leukaemia cells leading to the apoptosis of 90% of the cancerous cells over 72 hours (IC50 = 20uM). It has been suggested this is due to the reactivation of proapoptotic genes epigenetically repressed exclusively in cancer cells by SIRT1. In vitro, salermide has proved to be a more potent inhibitor of SIRT2 and overall is a stronger inhibitor than sirtinol.

5. Suramin Hexasodium Salt
Suramin hexasodium salt inhibits SIRT1, SIRT5, topoisomerase II and the cell surface binding of various growth factors. It is also a P2x and P2y purinergic receptor antagonist that displays antitumor, antiangiogenic and antiparasitic activities.

6. Tenovin-1
Tenovin-1 is a small molecule activator of p53 transcriptional activity. At 10µM, tenovin-1 elevates p53 expression in MCF-7 cells within two hours of treatment and longer-term exposure significantly decreases the growth of BL2 Burkitt’s lymphoma and ARN8 melanoma cells. Functioning upstream of p53, tenovin-1 acts by inhibiting the deacetylase activity of purified human SIRT1 and SIRT2, members of NAD+-dependent class III histone deacetylases that belong to the sirtuin family. While tenovin-1 demonstrates low genotoxicity, the compound has poor water solubility, which limits its uses in vivo.

7. Tenovin-6
Tenovin-6 is an analogue of Tenovin-1 that has been shown to reduce growth in ARN8 melanoma tumour grafts in SCID mice at 50mg/kg. The histone acetylase activity of SIRT1, SIRT2 and SIRT3 are also inhibited by Tenovin-6 at the following respective IC50 values 21µM, 10µM, & 67µM.
