Large Insert Library Construction
Cambridge Bioscience offers access to a library construction service for difficult-to-clone genomic DNAs, large cDNAs, and long PCR products from Lucigen. This service uses the first-of-its-kind BigEasy® pJAZZ® linear cloning vectors that represent a major advance in large insert library construction offering exceptional stability for large insert library construction.
Features of the BigEasy® pJAZZ® Linear Cloning Vectors
Traditionally, circular plasmid vectors are used for library construction whichoften become supercoiled in the host cell, causing torsional stress and insert instability.
pJAZZ vectors are best choice for constructing large insert libraries from difficult-to-clone genomic DNAs, large cDNAs and long PCR products due to the following features:
• Designed to have exceptional stability
• Remain linear
• Remain torsion-free
• Stably clones sequences unclonable in plasmid vectors
• Enables the construction of accurate and complete libraries, from <1 kb to over 30 kb
Deliverables
bSMART large insert libraries are delivered as frozen clones in 96- or 384-well plates, with a complete report and guidelines on storing and working with the bSMART Library.
Testimonial
“TIGR had tried several times [to make a large-insert Tetrahymena library], but the largest size range we could obtain was 4-6 kb. Lucigen was able to provide a library in the pJAZZ® vector with an average insert size of 12 kb and some inserts as large as 20 kb.”
Dr. Robert Coyne, The Institute for Genome Research (TIGR)
