Cambridge Bioscience offers access to a BAC shotgun sequencing service from Lucigen for the full length insert sequencing of large-insert cosmid, fosmid and BAC clones.

Method
Purified DNA from the cosmid, fosmid or BAC clone is randomly sheared, end-polished, size-selected, and subcloned into the exclusive pSMART® Vector from Lucigen and transformed into the E. cloni® 10G competent cells. The colonies of the shotgun library are robotically picked into 96-well plates for high-throughtput DNA preparation and Sanger sequencing. This service includes large-insert DNA preparation (optional), shotgun subcloning, library construction, colony picking, overnight culturing, DNA preparation from clones, DNA sequencing at any scale from a single sample to a 96-well high-throughput processing.
Data Provided
DNA sequences are transmitted via secure internet connection or provided on appropriate computer media on project completion. A complete shotgun library is provided at no additional cost.
