Biological Application of FRET – Protease Detection

In recent years, FRET based assays have found broad applications, one of which is the detection of protease activity. Proteases play key roles in protein activation, cell regulation and signaling, as well as in the generation of amino acids for protein synthesis or utilization in metabolic pathways.

FRET-based assays have made the detection of protease activity and High Throughput Screening of protease inhibitors faster and easier. AnaSpec, a world leader in FRET peptide technology, is proud to be the first in the industry to offer long wavelength protease assays for:

Cathepsins D, S
HCV (Hepatitis C Virus)
HIV
MMPs
α- (TACE), ß-Secretase (BACE1)
Renin
and WNV (West Nile Virus)

Heavy isotope labeled full-length human protein SRM/MRM Mass Spec Standards

In a "first of its kind" launch OriGene now offers the service to generate Mass Spectrometry (MS) standards for 5,000 human proteins. Produced in human HEK293T cells and labeled with [U- 13C6, 15N4]-L-Arginine and [U- 13C6, 15N2]-L-Lysine, the full length proteins with appropriate post-translational modifications are the best identification and quantification standards.

Heavy isotope labeled human proteins are ideal internal standards for SRM/MRM (single reaction monitoring, multiple reaction monitoring) mass spectrometry analyses for high throughput proteomics and clinical biomarker research.

  • Accurate quantification
  • Identify the best SRM and MRM transitions through experimental data
  • Authentic posttranslational modifications by using human HEK293T cell line.
  • Higher data consistency than synthetic peptide internal standard
  • Suitable for all types of MS equipment
  • Over 90% incorporation efficiency

Easy Effective Protein Cleavage Using Progenta AALS

The Progenta Anionic Acid Labile Surfactants (AALS) have been engineered to provide a safe, robust alternative to detergents (e.g. SDS) that are commonly used in proteomics work, but that negatively impact subsequent analysis by mass spectrometry.

While SDS and other detergents can improve protein solubility, they can be very difficult to remove during sample prep and purification of the protein sample. These bound detergents can cause significant impairment of protein analysis by mass spectrometry, as the surfactants can suppress analyte ion signal, promote analyte adduct formation, and present as contaminants during the analysis.

  • rapidly cleaved in just 10 mins at pH 2.5 – 3.0 in 1% TFA
  • cleavage products are fully soluble and do not interfere with mass spec
  • promotes protein solubility and improves enzymatic digestion
  • Certified Mass Spec Grade
