SBI's miRZip anti-microRNA Lentivectors

miRZip™ anti-sense microRNAs are stably expressed RNAi hairpins that have anti-microRNA activity. The miRZip shRNAs produce short, single-stranded anti-microRNAs that competitively bind their endogenous microRNA target and inhibit its function. The result is the elevation of the protein levels of the transcripts targeted by the microRNA being “zipped”.

The miRZip short hairpin RNAs are cloned into SBI’s pGreenPuro™ shRNA expression lentivector. The miRZip hairpins are rationally designed for asymmetry such that the upper strand of the hairpin (shown in gray in the attached image) does not contain the endogenous microRNA sequence and the lower strand is preferred for producing anti-sense microRNAs (shown in green in the attached image) that are fully complementary to a specific microRNA target.

HuSh 29-mer shRNA

HuSh 29-mer hairpin expression clones appear to enter the RNAi pathway more efficiently and result in much higher potency and specificity than shorter expressed or oligo RNAi forms. shRNA less than 30 base pairs in length evades the radar of the mammalian interferon response and initiates strong and specific gene silencing.
