Recombinant Human VEGF165
Recombinant proteins are predominantly produced in non-human cells such as E. coli and insect cells. The proteins produced by these methods lack authenticity due to the absence of physiologically relevant glycosylation. Furthermore, a number of important cytokines are not commercially available due to inadequate proteolytic processing, protein folding or other post-translational modifications that occur in the non-human cell expression systems.
Cambridge Bioscience offers VEGF165HuXp, an authentic recombinant human VEGF165 expressed in human 293 engineered cells from HumanZyme. This homodimeric glycoprotein's bioactivity was determined by its ability to induce the proliferation of human umbilical vein endothelial cells and subsequently has been shown to be 5 times more active than an E. coli expressed protein (ED50 of 1.6 ng/mL for HumaXpress™ expressed protein vs 7.9 ng/mL for E. coli expression).

Comparison of VEGF165HuXp & E. coli expressed VEGF165
E. coli expressed VEGF165 protein is a monomer - dimer mixture with a molecular mass of 18kD and 34kD when analysed by SDS-PAGE. This compares with the VEGF165HuXp which migrates as a glycosylated band of 36kD due to glycosylation and dimerization, illustrating the authenticity of VEGF165HuXp.
