Antibleaching Live Cell Visualization Medium
Photobleaching, often caused by oxidative reddening, of fluorescent proteins in response to prolonged exposure to exciting radiation significantly affects their utility as in vivo labels. In the past, the only known way to minimize photobleaching was to reduce the excitation light intensity or shorten the duration of illumination.
Cambridge Bioscience offers the antibleaching live cell visualization medium DMEMgfp from Evrogen designed to vastly increase the stability of GFP. This medium excludes the components that can cause oxidative reddening of green fluorescent proteins leading to up to a 9-fold increase of photostability of EGFP, a 3.3-fold increase of photostability of TagGFP2 and more than a 4-fold increase of photostability of activated forms of photoactivatable PA-GFP and PS-CFP2 (the effect varies for particular fusions and localizations).
Workflow
The use of DMEMgfp is as simple as replacing the standard cell culture medium with DMEMgfp 20-30 minutes before starting the imaging experiment under sterile conditions. This medium can be supplemented with L-glutamine, penicillin, streptomycin and fetal bovine serum. This medium is intended for imaging use and not for long term cell culture.
Benefits of Using DMEMgfp
• Up to 9-fold increase of photostability of EGFP
• Up to 3.3-fold increase of photostability of TagGFP2
• More than 4-fold increase of photostability of activated forms of photoactivatable PA-GFP and PS-CFP2
